Application of short hydrophobic elastin-like polypeptides for expression and purification of active proteins
Abstract To investigate the application of short elastin-like polypeptides (ELPs) in the purification of bioactive proteins, short hydrophobic ELP[I]n (n = 30, 40, 50) tags were constructed. Both the ELP[I]n tags and the ELP[I]n–Trx fusion proteins could be stably expressed in Escherichia coli and purified by inverse transition cycling, respectively. Total protein concentrations determined by BCA protein assay showed that the yield of the fusion proteins decreased with increasing...

Entrapment of enzyme in the presence of proline: effective approach to enhance activity and stability of horseradish peroxidase
Abstract In this report, activity and stability of horseradish peroxidase (HRP) entrapped in polyacrylamide gel in the presence of proline (HEP) are compared with that of enzyme entrapped in absence of proline (HE). Within polyacrylamide (8%) beads, 80% entrapment yield for peroxidase was observed in the presence as well as absence of proline. The HEP (1.5 M proline) showed 170% higher activity compared to HE. HEP also showed significant increase in KM, Vmax and Kcat. At 8th...

Priming maize seeds with cyanobacteria enhances seed vigour and plant growth in elite maize inbreds
Abstract Biofilm formation of a nitrogen-fixing cyanobacterium Anabaena torulosa with a beneficial fungus Trichoderma viride (An-Tr) was examined under laboratory conditions. A gradual enhancement in growth over A. torulosa alone was recorded in the biofilm, with 15–20% higher values in nitrogen fixation, IAA and exopolysaccharide production illustrating the synergism among the partners in the biofilm. To investigate the role of such biofilms in priming seed attributes, mesocosm...

A novel spectrophotometric method for simultaneous estimation of dibenzothiophene and 2-hydroxybiphenyl in their mixed spectrum and its application in screening of specific biodesulfurizing microbes
Abstract The desulfurization of fuel is currently enforced to meet environmental legislation and prevent pollution. The use of specific biodesulfurizing microbes with a unique 4S pathway allows the desulfurization without compromising the quality of fuel. These specific microbes can be screened by the detection of 2-hydroxybiphenol (2-HBP) in desulfurizing mixture of dibenzothiophene (DBT). At present, colorimetric Gibb’s assay is the most commonly employed screening method which...

Neuroblastoma SH-SY5Y cytotoxicity, anti-amyloidogenic activity and cyclooxygenase inhibition of Lasianthus trichophlebus (Rubiaceae)
Abstract The anti-amyloidogenic potential and cyclooxygenase anti-inflammatory activity of Lasianthus trichophlebus extracts were evaluated. The MeOH extract (LTM) and chloroform extract (LTC) exhibited significant cytotoxic inhibition against the neuroblastoma SH-SY5Y cell with an IC50 of 17.52 μg/mL and 12.28 μg/mL, respectively. Thioflavin T assay indicated the LTC extract inhibition (70.56% at 50 μg/mL) to be statistically comparable (p < 0.05) to the positive control....

Enhanced production of lipstatin from mutant of Streptomyces toxytricini and fed-batch strategies under submerged fermentation
Abstract Streptomyces toxytricini produces bioactive metabolite recognized as lipstatin and its intermediate orlistat. The main focus of this study is to enhance lipstatin production by strain improvement and precursor feeding. In this study, strain improvement to enhance the production of lipstatin was carried out by different doses (50, 100, 150, 200, and 250 Gy) of gamma radiation and precursors (Linoleic acid, Oleic acid, and l-Leucine). Screening showed that the highest...

Critical coronary artery ostial narrowing and sudden death
Abstract Coronary ostial stenosis is an uncommon lesion but one that may be associated with significant and potentially lethal reduction in distal artery blood flow. While most commonly due to atherosclerosis, it may also be caused by aortic arteritis, congenital webs or post-surgical complications following aortic valve replacement. Three cases of lethal ischemic heart disease are reported where occult ostial narrowing/occlusion was first identified at autopsy. Case 1: A 70-year-old...

Abstract The carcass of a 15-year-old female Bottlenose dolphin (Tursiops aduncus) was retrieved from the Port River near Adelaide, South Australia, Australia. The animal was emaciated with five thick nylon fishing lines emerging from the oropharynx attached to a tangle of nylon and monofilament fishing line that also contained wire and eight fishing hooks. Abstract We present the case of a 17-year-old man, who died after 2,4-dinitrophenol (DNP) and clenbuterol consumption, which he likely took for physical enhancement. Forensic post-mortem examination revealed a yellowish skin colour and nonspecific signs of asphyxia. Analytical confirmation of the intoxication was obtained in blood and urine, with high levels of DNP and clenbuterol. Both of these substances are used by bodybuilders as DNP enhance lipolysis and clenbuterol has anabolic...
