Increased thyroid uptake on 18F-FDG PET/CT is associated with the development of permanent hypothyroidism in stage IV melanoma patients treated with anti-PD-1 antibodies
Abstract Purpose To determine performances of 2-deoxy-2-(18F)fluoro-d-glucose (18F-FDG) positron emission tomography (PET) to detect the development of permanent thyroid dysfunction (PTD), and to evaluate the prognostic value of early increased thyroid uptake in stage IV melanoma patients treated with anti-programmed death 1 (anti-PD-1) antibodies. Methods ...

Synergistic effect of ibrutinib and CD19 CAR‐T cells on Raji cells in vivo and in vitro
Abstract Ibrutinib might improve the efficacy of anti‐CD19 chimeric antigen receptor (CD19 CAR) T cell therapy in chronic lymphocytic leukemia (CLL). We studied the possibility and mechanism of the synergistic effect of ibrutinib and CAR‐T cells in other types of lymphoma. In this study, we selected the CD19 CAR‐T cells of a patient with lymphoma who failed in his CD19 CAR‐T cell therapy and a dose of 8 mg/kg/day ibrutinib. Subcutaneous and tail vein tumorigenic mice were established with Raji cells....

Complex roles of the actin‐binding protein Girdin/GIV in DNA damage‐induced apoptosis of cancer cells
Abstract The actin‐binding protein Girdin is a hub protein that interacts with multiple proteins to regulate motility and Akt and trimeric G protein signaling in cancer cells. Girdin expression correlates with poor outcomes in multiple human cancers. However, those findings are not universal, as they depend on study conditions. Those data suggest that multiple aspects of Girdin function and its role in tumor cell responses to anticancer therapeutics must be reconsidered. In the present study, we...

Comprehensive genomic profiling of gastroenteropancreatic neuroendocrine neoplasms (GEP-NENs)
Purpose: GEP-NENs are rare malignancies with increasing incidence. Their molecular characteristics are still undefined. We explored the underlying biology of GEP-NENs and the differences between gastrointestinal (GI) and pancreatic (PNET), high grade (HG) and low grade (LG) tumors. Experimental Design: GEP-NENs were analyzed using next-generation sequencing (NGS; MiSeq on 47 genes, NextSeq on 592 genes), immunohistochemistry, and in-situ hybridization. Tumor mutational burden (TMB) was calculated...

Targeting isocitrate dehydrogenase mutations in cancer: emerging evidence and diverging strategies
Isocitrate dehydrogenase active-site mutations cause a neomorphic enzyme activity that results in the formation of supraphysiological concentrations of D-2-hydroxyglutarate (D-2HG). D-2HG is thought to be an oncometabolite that drives the formation of cancers in a variety of tissue types by altering the epigenetic state of progenitor cells by inhibiting enzymes involved in histone and DNA demethylation. This model has led to the development of pharmacological inhibitors of mutant IDH activity for...

Assessing the Impact of a Patient Navigator Intervention Program for Vietnamese-American Women with Abnormal Mammograms
Abstract Breast cancer incidence among Asian Americans increased at a rate of 1.8% per year from 2014 to 2018, while the general population’s incidence rate remained stable. Vietnamese-American women have been found to have the longest follow-up time after an abnormal mammogram. This study assesses the impact of a patient navigator program for Vietnamese-American women who received abnormal mammograms. Ninety-six Vietnamese-American participants with abnormal mammograms were assigned...

‘Steeling’ Effects in the Association between Psychological Resilience and Cancer Treatment in Prostate Cancer patients
Abstract ObjectiveTo test the ‘buffering’ effect of Psychological Resilience upon depression in prostate cancer patients, and to also investigate any effects that past or current treatment may have had upon patients’ Psychological Resilience as a test of the ‘steeling’ hypothesis of past adversity upon future resilience.MethodsA total of 576 volunteer prostate cancer patients completed questionnaires about their demographic and treatment variables, and their Psychological Resilience and depression....
