Absolute Quantification of Cell-Free Protein Synthesis Metabolism by Reversed-Phase Liquid Chromatography-Mass Spectrometry
Here, we present a robust protocol to quantify 40 compounds involved in central carbon and energy metabolism in cell-free protein synthesis reactions. The cell-free synthesis mixture is derivatized with aniline for effective separation using reversed-phase liquid chromatography and then quantified by mass spectrometry using isotopically labelled internal standards.

Experimental Approach to Examine Leptin Signaling in the Carotid Bodies and its Effects on Control of Breathing
Our study focuses on the effects of leptin signaling in carotid body (CB) on the hypoxic ventilatory response (HVR). We performed 'loss of function' experiments measuring the effect of leptin on HVR after CB denervation and 'gain of function' experiments measuring HVR after overexpression of the leptin receptor in CB.

Site‐Selective Alkoxylation of Benzylic C–H Bonds via Photoredox Catalysis
Methods that enable the direct C–H alkoxylation of complex organic molecules are significantly underdeveloped, particularly in comparison to analogous strategies for C–N and C–C bond formation. In particular, almost all methods for the incorporation of alcohols via C–H oxidation require the use of the alcohol component as solvent. This limits the practical scope of these reactions to simple, inexpensive alcohols. We report a photocatalytic protocol for the functionalization of benzylic C–H bonds...

Ultra‐Stable Plasmonic Colloidal Aggregates for Accurate and Reproducible Quantitative SE(R)RS in Protein‐Rich Biomedia
Au/Ag colloids aggregated with simple salts are amongst the most commonly used substrates in surface‐enhanced (resonance) Raman spectroscopy (SE(R)RS). However, salt‐induced aggregation is a dynamic process, which means that SE(R)RS enhancements vary with time and that measurements therefore need to be taken at a fixed time point, normally within a short time‐window of a few minutes. Here, we present an emulsion templated method which allows formation of densely‐packed quasi‐spherical Au/Ag colloidal...

X‐ray Crystallography and Vibrational Spectroscopy Reveal the Key Determinants of Biocatalytic Dihydrogen Cycling by [NiFe] Hydrogenases
Where does the H2 go? Using an F420‐reducing [NiFe]‐hydrogenase from Methanosarcina barkeri as a model enzyme, it is shown that the protein backbone provides a strained chelating scaffold that tunes the [NiFe] active site for efficient H2 binding and conversion. The protein matrix also directs H2 diffusion to the [NiFe] site via two gas channels and allows the distribution of electrons between functional protomers through a subunit‐bridging FeS cluster. An Aptamer‐Nanotrain Assembled from 6‐Letter DNA Delivers Doxorubicin Selectively to Liver Cancer Cells
Expanding the number of nucleotides in DNA increases the information density of functional DNA molecules, creating nanoassemblies that cannot be invaded by natural DNA/RNA in complex biological systems. Here, we show how 6‐letter GACTZP DNA contributes this property in two parts of a nanoassembly: (1) in an aptamer evolved from a 6‐letter DNA library to selectively bind liver cancer cells; and (2) in a 6‐letter self‐assembling GACTZP nanotrain that carries the drug doxorubicin. The Anaphase‐Promoting Complex: a key mitotic regulator associated with somatic mutations occurring in cancer
Abstract The anaphase‐promoting complex/cyclosome (APC/C) is an E3 ubiquitin ligase that helps control chromosome separation and exit from mitosis in many different kinds of organisms, including yeast, flies, worms, and humans. This review represents a new perspective on the connection between APC/C subunit mutations and cancer. The complex nature of APC/C and limited mutation analysis of its subunits has made it difficult to determine the relationship of each subunit to cancer. In this work, cancer...

Testing NTRK testing: wet‐lab and in silico comparison of RNA‐based targeted sequencing assays
Abstract NTRK fusions involving the three neurotrophic tyrosine receptor kinase genes NTRK1, NTRK2, or NTRK3, and a variety of fusion partners were identified as oncogenic drivers across many cancer types. Drugs that target the chimeric protein product require the identification of the underlying gene fusion. This advocates the diagnostic use of molecular assays ranging from fluorescence in‐situ hybridization (FISH) and RT‐PCR/Sanger approaches to targeted next generation sequencing.
